<概要/目的>
To improve our ability in explaining our research and enhance our insight not only in our research topic but also in different field. Through this program, we aim to develop a big chance to each other to practice our communication and discussion skill through different language. So, we can learn from each other how to get and give better understanding about our research topics.

<方法/企画としての特徴>
Have one UOS student to stay in our laboratory, do discussion each other about research and experiment that we are doing and the goals that we are going to achieve. Moreover, doing some basic experiment will be beneficial to gain better knowledge and get better understanding in research that we were discuss.

<活動内容/具体的成果>
Day 1.
1.        Introduction
I introduced one UOS student, Roh Hyun Soo to my laboratory member. In the laboratory, we took a rest for a while and then I gave some explanation to him about our basic work in laboratory and the implementation for many research topics in our field.
2.        Tour to Green House
I invited him to the green house to show the rice plant that we usually use in laboratory and show how to select the best rice flower for doing experiment to get good quality of rice sperm and rice egg cells.

Day 2
1.        Isolation of the rice ovaries
We went to green house in the morning and pic some rice flower. After pic the rice flower I explained him how to break the flower and isolate the ovary by using stereo microscope. He took quite long time to isolate the ovary, since removing the stamen and the stigma from the ovary seems to be difficult for him. We keep the rice ovary in 4°C room temperature to be use for the next day.
2.        Welcoming party
In the evening I and laboratory member held a welcoming party to Hyunsoo, we prepared some Japanese food and drink during the party. We were talked a lot and sharing about our each other’s experiment in our country, not only about research but also about life and environment.

 
Day 3
1.        Preparation of cover glass
To put our sample (rice gametes and zygotes) safely, we need the cover glass which bordered with silicon. I tough him how to make silicon border around the cover glass by using diethyl ether in the fume hood. It was very easy, so that he could do it very well.
2.        Isolation of the rice egg cells
We used the rice ovary which we isolated from previous day to isolate the egg cells. I explained to him and gave an example how to cut the ovary in correct position, so we can get good and a lot o of ovary. He could do it very well and successfully isolate about six egg cells only in short time and transfer them to mannitol droplets on the cover glass.
3.        Isolation of sperm cells
I explained him how to isolate the sperm cells inside the pollen grain from the anther by using forceps to break the anther inside the mannitol solution. Since the sperm cells was very small and transparent, I showed him how to isolate the sperm cells under inverted microscope which connected to the camera and PC with Instudio software, so then he could see it from the computer screen. In this experiment, he needed quite long time to find and isolate the sperm cells, but finally he could do it.
4.        IVF System (electrofusion)
By using the isolated rice egg cells and the sperm cells, I showed him how to produce the IVF rice zygotes through electrofusion. For the first time I did the electrofusion and he observed it trough computer screen, after that he tried it by himself. He was very talent, because in the first time he learns, he could get 3 IVF rice zygotes.


Day 4
1.        Zygotes culture
In the last day, I tough him how to prepare the culture medium to culture the rice zygotes so that we could produce the whole rice plant. But, at that time we did not have enough time to do electrofusion anymore, so we just did the simulation of the zygotes culture.


<感想/課題など(企画力/評価力/自主性等の向上を含む)>
私は主に、イネの卵細胞・精細胞の単利の方法と、それらの細胞を用いた細胞融合のやり方を教えた。細胞の単離は顕微鏡の扱い方や、卵細胞の特徴や見方が理解できないと困難な作業であるため、あらかじめ文章でプロトコルを作成したり、画像を用いることでスムーズに理解してもらおうと努めた。また、研究室内での交流を深めるために、研究室内での食事会を企画・運営にも携わりました。お互いの国の文化や、研究の話をフランクに話し合えたので、自身にとってもラボのメンバーにとっても有意義な時間になったと感じています。(M2 出牛 稜也)
